La respuesta hipofisaria al GnRH depende del número de receptores, que aumenta después del nacimiento y sufre variaciones cíclicas durante el ciclo reproductor y está modulada por esteroides, neurotransmisores, insulina, opioides, etc. La administración prolongada de un agonista del GnRH (esteroides, por ejemplo) produce fallos en la esteroiodogénesis y espermatogénesis. En el macho, los andrógenos inhiben fundamentalmente la secreción de LH.
